Nylock nuts
Instead of Locktite I bought a couple boxes of nylock nuts. Easier to use and less messy.

Amen to that!
Just remember that the Nylon part doesn't last forever.
Always inspect a nylock nut before reusing it.
Also, a Nylock nut needs a longer screw than a normal nut, so you may need to replace some with longer ones.

I use nylocs with anti vibration washers to clamp my router mounts. Perfect for the job but occassional dissasembly (to use the router by hand) is a PITA with the nyloc resisting all the way. Still better than cleaning up and reapplying loctite at these particular nuts and bolts in my opinion and since my mounts are HDPE I wanted to be sure I wasn't going to chemically attack them with loctite. I think there is a case for threadlock in places and a case for nylocs and even anti vibration washers if that's what you want to do, I think of these as minor mods to suit individual preference or as a knock effect of a more major mod.

What sizes (and number per size) will I need for a complete build?

That really depends...
For M5 nuts, you need 3 x 4 for the V-wheels, 2 x 3 for idlers. That's 18.
(That was for my reasonably stock ShapeOko. Not certain about SO2)
Depending on your model SO and adaptations, that may be enough...
But if they're sold in boxes of 20, I'd get one extra just in case.
There's a lot of M5 bolts that aren't held by a nut, after all.
